Abstract

Aktivitasenzimatikdari metabolit ekstraseluler bakteri dapat dikembangkan sebagai kemoterapi antikanker. Terdapat tiga golongan toksin berdasarkan aktivitas enzimatiknya, yaitu hidrolitik, transferase dan lyase, serta golongan cytolysin. Streptococcus pyogenes menghasilkan deoxyribonuclease sebagai enzim hidrolitik, NAD-glycohydrolase sebagai transferase dan lyase, serta streptolysin O dan streptolysin S sebagai cytolysin. Penelitian ini bertujuan untuk menguji efek sitotoksik dari supernatan medium kultur Streptococcus pyogenes dalam menginduksi kematian sel HeLa. Kultur sel kanker serviks (sel HeLa) diberikan perlakuan dengan variabel dosis supernatan medium kultur bakteri, yaitu 0% (kontrol), 9,09%, 16,67%, dan 28,57%, serta variabel lama inkubasi pemaparan, yaitu 120 dan 60 menit. Kemudian dilakukan uji viabilitas sel dengan MTT Assay untuk mengukur survival rate dari sel HeLa berdasarkan tingkat absorbansi spektrofotometri dan dilanjutkan uji statistik dengan program SPSS. Pada uji One-Way Anova quadratic term didapatkan perbedaan yang signifikan p = 0,021 (p < 0,05) untuk inkubasi 120 menit, dan p = 0,023 untuk inkubasi 60 menit. Sedangkan pada uji regresi kuadratik didapatkan R = 0,722; p = 0,036 untuk inkubasi 120 menit, dan R = 0,705; p = 0,045 untuk inkubasi 60 menit. Pada analisis t-test antara kelompok inkubasi 120 menit dengan 60 menit didapatkan nilai p dari masing-masing dosis (9,09%, 16,67%, dan 28,57%) secara berturut-turut adalah p = 0,058; p = 0,787; dan p = 0,660 (p > 0,05). Jadi, dapat disimpulkan bahwa terdapat perbedaan cells survival rate yang bermakna di antara masing-masing dosis dengan pola penurunan cells survival rate secara signifikan sesuai dengan model regresi kuadratik, tetapi tidak ada perbedaan yang bermakna antara kelompok inkubasi 120 menit dengan 60 menit. Penelitian lebih lanjut diperlukan mengenai aktivitas sitotoksik strain bakteri spesifik, respon target sel kanker yang lain, dan waktu inkubasi yang lebih mendetail.

English Abstract

Enzymaticactivityof extracellular bacterial metabolites could be developed as anticancer chemotherapy. There are three classes of toxins based on the enzymatic activity, such as hydrolytic, transferase and lyase, and cytolysin. Streptococcus pyogenes produces deoxyribonuclease as hydrolytic enzyme, NAD-glycohydrolase as transferase and lyase, and streptolysin O and streptolysin S as the cytolysin. The purpose of this study is to prove the cytotoxic effect of Streptococcus pyogenes’ medium culture supernatant in inducing the death of HeLa cells. The cells culture of cervical cancer (HeLa cells) are given treatments by the variable doses of bacteria culture medium supernatant consist of 0% (control), 9.09%, 16.67%, and 28,57%, as well as the variable of exposing incubation time that are 120 and 60 minutes. Then cells viability test is conducted with MTT Assay to measure the survival rate of HeLa cells based on absorbance rate of spectrophotometry and continued with the statistic analysis by SPSS program. From the One-Way Anova by quadratic term obtained significant difference p = 0,021 (p < 0.05) for 120 minutes incubation, and p = 0,023 for 60 minutes incubation. From the quadratic regression test obtained R = 0,722; p = 0,036 for 120 minutes incubation, and R = 0,705; p = 0,045 for 60 minutes incubation. Whereas from Independent Samples t-test between 120 minutes group and 60 minutes group obtained p value from each doses (9,09%, 16.67%, and 28,57%) respectively are p = 0,058; p = 0,787; and p = 0,660 (p > 0.05). So, it can be concluded that there is a significant difference of cells survival rate between each dose treatment with a pattern of decreasing cells survival rate which appropriates significantly with quadratic regression model, but there is no meaningful difference between 120 minutes-incubation group and 60 minutes-incubation group. More advance researches are needed regarding the cytotoxic activity of specific bacterial strains, response to other cancer cell targets, and more detailed incubation time.
