Abstract

Genistein merupakan senyawa yang banyak dalam kedelai, efek Genistein yang telah lama dikenal adalah inhibitor tirosin kinase, sehingga dapat menghambat ekspresi VEGFR. VEGF berfungsi dari awal vaskulogenesis, blood islands, kapiler dan morfologi jantung. Penelitian ini bertujuan untuk mengetahui pengaruh pemberian Genistein terhadap vaskulogenesis dan angiogenesis embrio, dengan model coba embrio ayam umur 48 jam. Penelitian ini merupakan penelitian analitik eksperimental, yang memiliki jumlah sampel 7 embrio masing-masing kelompok perlakuan, sesuai dengan rumus Steel dan Torrie. Variabel yang diukur adalah survival rate, jumlah somit, tahapan pembentukan pembuluh darah, dan persentase luas area VEGFR-2. Prosedur penelitian adalah dengan menginjeksi telur dengan larutan Genistein, menginkubasi selama 48 jam, lalu mengamati hasilnya dengan pengecatan imunohistokimia dan sterologi. Hasil Penelitian menunjukkan bahwa Genistein tidak secara signifikan berpengaruh terhadap survival rate embrio ayam (p≥0.05) dan jumlah somit (p≥0.05). Namun menunjukkan perbedaan yang bermakna terhadap tahapan pembentukan pembuluh darah dan ekspresi VEGFR-2 (p≤0.05). Hasil penelitian menunjukkan hasil dengan kecenderungan yang sama yaitu menurun dan meningkat lagi pada dosis tinggi, walaupun survival rate dan jumlah somit tidak menunjukkan hasil yang signifikan. Peningkatan dosis tertinggi (40µM) pada hitung Persentase area VEGFR-2 tidak menunjukkan peningkatan yang bermakna terhadap dosis sebelumnya (20µ). Kesamaan pola ini belum diketahui mekanismenya. Kesimpulan dari penelitian ini adalah Genistein menghambat vaskulogenesis dan angiogenesis embrio ayam umur 48 jam, namun tidak sampai menimbulkan kematian embrio dan gangguan morfologi embrio (melalui perhitungan somit) secara signifikan.

English Abstract

Genistein is a substance rich in soy. It has effect that has been long known as tyrosine kinase inhibitor, so it can inhibit VEGFR expression. VEGF work from early vasculogenesis, blood islands, capillary, and heart formation. This research is to know the effect of Genistein toward vasculogenesis and angiogenesison embryo, with chicken embryo as the research model. This is an analytic experimental research which has 7 samples on each group, following the formula from Steel and Torrie. Variable measured at this research are: survival rate, vascular formation, somite count, and Area Persentation of VEGFR-2 expression. This research procedure include the injection of Genistein to the yolk, incubation within 48 hours, then observation with immunohistochemistry and sterology procedure. Results of this research is that Genistein not significantly effect the survival rate and somite count of the 48 hours aged chicken embryo (p≥0.05). Genistein have significant differences within control and experiment group of vascular formation and area persentation of VEGFR-2 expression (p≤0.05).The results of this research has the same trend, that it decreases then increases again at high dose, altough results of somite count and survival rate did not give significant results. Increased VEGFR-2 expression on highest dose (40µM) did not give significant increase toward the dose befor it(20µ). The mechanism of this same pattern has not yet been known. This research conclude that Genistein has effect toward vasculogenesis and angiogenesis of 48 hours aged chicken embryo, but does not cause embrionic death and morphology interruption significantly.
