Abstract
Salmonella Typhi adalah salah satu patogen penyebab penyakit demam tifoid. S. Typhi dilaporkan sudah resisten terhadap beberapa obat antimikroba sehingga perlu dilakukan penelitian untuk menemukan alternatif terapi lain. Salah satu alternatif terapi adalah menggunakan bahan alami yaitu rimpang lengkuas merah (Alpinia purpurata). Kandungan aktif yang diduga bermanfaat sebagai antimikroba adalah flavonoid, terpenoid, dan minyak atsiri. Penelitian ini bertujuan untuk mengetahui potensi antimikroba ekstrak etanol rimpang lengkuas merah terhadap S. Typhi. Penelitian ini merupakan penelitian eksperimental laboratorik dengan menggunakan metode dilusi agar untuk menentukan KHM. Sampel yang digunakan dalam penelitian ini adalah 1 isolat S. Typhi yang diperoleh dari Laboratorium Kesehatan Yogyakarta. Konsentrasi akhir ekstrak etanol rimpang lengkuas merah yang digunakan yaitu 0,1%, 0,2%, 0,3%, 0,4%, 0,5% dan 0,6% v/v dengan empat kali pengulangan, sedangkan konsentrasi S. Typhi adalah 106 CFU/ml. Hasil uji statistik menunjukkan bahwa ekstrak etanol rimpang lengkuas merah, secara signifikan dapat menghambat pertumbuhan S. Typhi (Kruskal Wallis, p =0.000) dan terdapat hubungan antara peningkatan konsentrasi ekstrak etanol rimpang lengkuas merah dengan penurunan jumlah pertumbuhan koloni S. Typhi (Spearman, r = -0.964). KHM ekstrak etanol rimpang lengkuas merah terhadap S. Typhi adalah 0.5% v/v. Dengan demikian, dapat disimpulkan bahwa ekstrak etanol rimpang lengkuas merah memiliki efek antimikroba terhadap S. Typhi secara in vitro dengan KHM 0,5% v/v.
English Abstract
Salmonella Typhi is a pathogen causing typhoid fever. S. Typhi is reported resistant towards several antimicrobial drugs, so it’s needed to find other alternative therapy. Alternative therapy is the use of natural materials the Red Galangal Rhizome (Alpinia purpurata). The active substances suspected having antimicrobial effect are flavonoids, terpenoids, and essential oils. This experiment aims to verify the antimicrobial effects of ethanolic extract of Red Galangal Rhizome against S. Typhi. The antimicrobial test used in this experiment is the agar dilution test to determine the Minimum Inhibitory concentration (MIC). The sample used in this study were 1 isolates of S. Typhi obtained from the Laboratory of Medicine, Yogyakarta. The final extract concentration used were 0.1%, 0.2%, 0.3%, 0.4%, 0.5% and 0.6% v/v , with four repetitions, while the concentration of S. Typhi was 106 CFU/ ml. Statistical analysis showed that the ethanolic extract of Red Galangal Rhizome could significantly inhibit the growth of S. Typhi (Kruskal Wallis, p = 0.000) and there was a relationship between the increase ethanolic extract of Red Galangal Rhizome concentration with the decrease of S. Typhi colony numbers (Spearman, r = -0.964). The research also showed that the MIC was 0.5% v/v. It can be concluded that ethanolic extract of Red Galangal Rhizome has an antimicroba effect against S. Typhi in vitro with MIC was 0,5% v/v.
